Jaysh al Islam, which has up to 20,000 trained fighters and controls suburbs east of the capital, was recently represented at landmark opposition talks in Saudi Arabia.
The Syrian Observatory for Human Rights confirmed five other Jaysh al Islam commanders were killed "in an airstrike that targeted one of their meetings in Eastern Ghouta".
At least six more rebel fighters were killed.
Alloush's death "stands as one of the most significant opposition losses" of Syria's nearly five-year uprising, analyst Charles Lister said on Twitter.
It comes amid a huge Syrian operation to retake rebel-held Eastern Ghouta.
President Bashar al-Assad's forces are backed by Russia, whose air force has completed 5,240 sorties since joining the conflict in September.
Footage has emerged of some of their latest strikes in which Islamic State targets including oil facilities were apparently hit.
